  • Working with the new real-time PCR assay launched by Novacyt called Genesig. This machine is a CE-marked three-gene assay used to detect the gene targets of SARS-CoV-2.
  • It is important and crucial that QC are carried out to verify the validity of results. The assay includes an internal control to identify possible PCR inhibition, measure extraction purity and confirm the integrity of the run. My role involves using pos/neg controls. The positive control allows us to be sure that the experimental set-up is working correctly. This increases the reliability of the experiment and testing against these predetermined specifications ensures consistent product quality.
  • The negative control is called the ‘baseline’. These controls are maintained in experiments to eliminate experimental errors and biased results. After the run has been completed a sigmodial curve graph is produced, which enables us to analyse whether the sample has reached the positive or negative threshold. The Positive control should produce a Cq value (quantification cycle) between 14-22 in the FAM channel. The negative control should produce a Cq value of less than 30 in the HEX channel.
